愈来愈多的基因被报道参与到水稻的缺磷信号转导网络中,但参与该网络的WRKY家族转录因子还鲜有报道。为了研究水稻WRKY转录因子在缺磷胁迫响应中的功能及其调控机制,本工作从水稻中克隆到一个在转录水平响应缺磷信号的WRKY转录因子OsWRKY-P1。通过转基因手段和RT-qPCR技术,对OsWRKY-P1的亚细胞定位情况及其过量表达转基因植株的生理参数及下游缺磷诱导表达基因的表达的变化情况进行了分析。结果显示,过量表达OsWRKY-P1导致正常供磷转基因植株叶片中的可溶性磷含量提高,但根部磷含量不变。有趣的是,除PHT1家族磷转运蛋白的一个低丰度成员,即OsPHT1;12的表达发生上调外,OsWRKY-P1过量表达并未导致其他PHT1成员表达量的增加。这些结果表明,OsWRKY-P1参与维持水稻磷素的动态平衡,但其分子机制有待进一步深入的工作进行解析。

n increasing number of genes have been reported to play pivotal roles in the phosphate starvation signaling network in rice. However, few attentions have been paid to rice WRKY transcription factors involved in this network. In order to elucidate the functions and regulatory mechanisms of rice WRKY transription factors in phosphate starvation responses, a phosphate starvation-responsive WRKY transcription factor, OsWRKY-P1, was identified in this work. By transgenic approaches and RT-qPCR analysis, the subcellular localization, physiological parameters and the regulation of the downstream phosphate starvation-induced genes were investigated in the overexpression lines. The results showed that both the soluble phosphate concentration was increased in the Pi-replete OsWRKY-P1 overexpression lines as compared with the wild-type plants. Unexpectedly, only one phosphate transporter gene of the PHT1 family, OsPHT1;12, which has a extremely low expression level, was upregulated upon OsWRKY-P1 overexpression. Taken together, OsWRKY-P1 is involved in phosphate starvation responses, and the underlying molecular mechanisms need to be further explored.
