激素配比对白桦悬浮细胞中三萜积累的影响

以白桦 (Betula platyphylla Suk.) 组培苗诱导的愈伤组织为研究材料,在悬浮培养的一个周期内,IS培养基上附加不同种类和浓度的激素,观察不同激素组合对白桦悬浮细胞生长及其三萜积累的变化。结果表明:在白桦细胞悬浮培养的一个周期内,悬浮细胞的活力和其内的三萜含量均于第9d达到最大值,分别为1.42 和14.31 mg•g-1 (DW),由此建议悬浮细胞每7~10 d更换一次培养液。在6-BA与TDZ和6-BA与NAA激素组合中,6-BA与TDZ组合最有利于白桦悬浮细胞干重和三萜的积累。其中,0.6 mg•L-1 TDZ +0.2 mg•L-1 6-BA下最有利于悬浮培养细胞干重和三萜产量的积累,收获时的干重和三萜产量分别为0.99 g•瓶-1和 0.22 g•L-1(DW);而0.4 mg•L-1 6-BA +0.2 mg•L-1TDZ培养的悬浮细胞中的三萜含量最高,收获时的三萜含量和产量分别达到14.23mg•g-1(DW)和0.21g•L-1(DW)。由此可见,有利于白桦三萜积累的激素组合和浓度为0.4 mg•L-1 6-BA +0.2 mg•L-1TDZ。

allus were induced by birch stems on IS medium and suspension cells were used to study the effect of the types and concentrations of plant hormones on the relation between the growth situation and the content of triterpenoids. During one suspension culture cycle of birch, the cell viability and triterpenoid content of suspension cell reached the maximum after 9 days, and the values were 1.42 and 14.31 mg • g-1 (DW), respectively. These results suggest that suspension culture liquid replacement was every 7~10 d. On the hormone combinations between 6-BA + NAA and 6-BA + TDZ, 6-BA与TDZ were beneficial to accumulation of cell dry weight and triterpenoids. Among of them, 0.6mg•L-1 TDZ +0.2 mg•L-1 6-BA was best to increase of dry cell weight and triterpenoid yield, the suspension cell weight and triterpenoid yield cultured after 25d was 0.99 g•bottle-1 and 0.22 g•L-1, respectively; 0.4 mg•L-1 6-BA +0.2 mg•L-1TDZ was best to accumulation of triterpenoids, the content and yiled of triterpenoids cultured after 25d was 14.23mg•g-1and 0.21 g•L-1, respectively. It was concluded from above results that the best culture condintion for triterpenoid accumulation was 0.4 mg•L-1 6-BA +0.2 mg•L-1TDZ.
