目的:探究甘露糖对结直肠肿瘤发生及增殖等影响,初步探究影响机制。建立原发结直肠癌小鼠模型,发现甘露糖的干预,能够显著降低结直肠肿瘤病灶的发生率以及演进程度;细胞水平实验,通过MTT实验,发现一定剂量甘露糖可以显著减弱结直肠癌细胞系HCT116及HT29的增殖,且在刺激浓度为25 mmol/L,作用48 h后,能够显著性降低两者培养液中葡萄糖的消耗量,分别达到68.40%及58.78%;通过蛋白质印迹法,发现甘露糖可降低这两株细胞系中葡萄糖转运载体蛋白GLUT1表达,在动物模型的结直肠组织中也得到了一致的结果。结论:甘露糖能够在体外干扰结直肠癌细胞葡萄糖摄取及增殖,降低原发结直肠肿瘤小鼠结直肠肿瘤发生率及演进程度,推测此现象与葡萄糖转运载体GLUT1表达下调有关。

Purpose: to explore the effect of mannose on colorectal tumorigenesis and proliferation, and the mechanism. The mouse model of primary colorectal cancer was established. The intervention of mannose could significantly reduce the incidence and progression of colorectal tumor. In the vitro experiment, through the MTT experiment, it was found that mannose could significantly reduce the proliferation of colorectal cancer cell lines HCT116 and HT29, and after the stimulation concentration of 25 mmol/L, for 48 hours, it could significantly reduce the glucose consumption in the culture medium of the two cell lines, reaching 68.40% and 58.78%, respectively. By Western blotting, it was found that mannose could reduce the expression of glucose transporter protein GLUT1 in these two cell lines, and the same results were obtained in vivo experiment. Conclusion: mannose can interfere with glucose uptake and proliferation of colorectal cancer cells in vitro and reduce the incidence and progression of primary colorectal tumor in mice. It is speculated that this phenomenon is related to the down-regulation of glucose transporter GLUT1 expression.
