pH响应型巯基肽特异富集材料壳聚糖衍生物的合成优化及应用
Synthesis optimization of chitosan derivatives containing 5-MP group and application in selection of cysteine containing peptides based on pH response
在蛋白组学分析中,如何实现巯基肽的高效、特异富集一直是一个挑战.针对该问题,本研究首先合成了具有pH响应自组装/解自组装和巯基捕获/释放功能的壳聚糖衍生物的富集材料O-CMC-PBS-5MP,并通过红外光谱分析、全波长扫描分析、核磁共振碳谱对合成的富集材料进行表征;然后探索模拟pH响应条件下壳聚糖分子量对接枝5MP的稳定性影响;接着,以巯基肽GC-7作为捕获对象探究了该材料捕获/释放巯基肽的条件,并以高丰度非巯基8肽作为干扰,验证O-CMC-PBS-5MP对含巯基肽段的捕获特异性;最后探索将特异富集的巯基肽直接应用于LC-MS和MALDI-MS分析可行性.结果表明,O-CMC-PBS-5MP在酸性条件下发生自组装絮凝并沉淀,而沉淀在碱性条件(pH≥10.0)下又复溶,以400 KDa壳聚糖为载体的5MP产物O-CMC-PBS-5MP,其5MP在整个过程中最为稳定,其对巯基肽GC-7的捕获率可达到90%,释放率达到54%左右,对巯基肽GC-7的特异性富集率达50%,并且富集的产物可以直接应用于LC-MS和MALDI-MS分析。?????
How to realize efficient and specific enrichment of cysteine containing peptides from tryptic digests of complex protein mixtures has always been a challenge. Firstly, this paper describes the development of chitosan derivatives (O-CMC-PBS-5MP) with the ability of self-assembly and capture / release of cysteine containing peptides based on pH responsive, and then the synthesized enrichment material was characterized by infrared spectroscopy, full wavelength scanning analysis and 13C-NMR. The effect of chitosan molecular weight on the stability of covalent 5MP group treated with simulated pH condition was expolred. Furthermore, the capture ability specificity of O-CMC-PBS-5MP for cysteine containing peptides was tested using GC-7 containing cysteine, as well as, the capture specificity of O-CMC-PBS-5MP for cysteine containing peptides was done using GC7 and 8 peptide without cysteine as the interference in high ratio. Finally, the selected cysteine containing peptides of GC7 from the mixture of GC7 and 8 peptide in high ratio was used directly for LC-MS and MALDI-TOF-MS analysis. The results of these studies showed that O-CMC-PBS-5MP is precipitated under acidic conditions (pH<3) and the precipitation was re-dissolved under alkaline conditions (pH ≥ 10.0). The 5MP group of O-CMC-PBS-5MP with 400 kDa chitons was the most stable in the whole simulated selection process, which capture rate of O-CMC-PBS-5MP for GC-7 could reach 90% and the release rate could reach approximately 54%, and the specific enrichment rate of mercaptopeptide GC-7 could reach 50%. The selected GC7, which will not need further treatment, could be directly applied for the LC - MS and MALDI - MS analysis.
