利用小RNA深度测序鉴定锦紫苏类病毒

类病毒是一类可以侵染高等植物的特殊病原体,在特定的宿主中可以影响其正常的生理、生化功能和生长发育过程,并可以引起植物发病。应用高通量测序技术对植物的小RNA进行深度测序和生物信息学分析,从而鉴定受试植物感染的已知类病毒和发现新类病毒是本实验室发展并应用的类病毒研究新方法。本论文利用深度测序技术对观赏花卉锦紫苏的小RNA进行分析,对小RNA文库进行PFOR组装和BLASTN分析,发现已知类病毒Coleus blumei 1和Coleus blumei 5。另外还发现一条序列与数据库中的序列无任何序列相似性,但该RNA的环状特性和碱基组成具有类病毒的典型特征,因此推测该条序列可能为新的类病毒序列。
Viroids are the infectious pathogens of plants that affect the physiological and biochemical process and normal development of certain hosts thereby causing diseases. Application of high-throughput sequencing of small RNAs isolated from infected plants and bioinformatic analysis using PFOR algorithm first developed and applied by our laboratory discovered and identified known and unknown viroids. In this study, the main objective is to discover viroid in an ornamental plant named Coleus blumei combining small RNA sequencing and bioinformatic analysis. Assembly by PFOR and BLASTN at nucletide level led to the discovery of two known viroids named Coleus blumei viroid 1 and Coleus blumei viroid 5. Another circular RNA sharing no sequence similarity to any of the known viroids was hypothesized as a new viroid based on its circular nature and the composition of nucleic acid.
