高温胁迫对高羊茅细胞膜及其核DNA伤害的初步研究

通过盆栽试验研究了高温逆境(35/30°C,白/昼,20d)对两种耐热性不同的高羊茅(热耐受型美洲虎3号和热敏感型TF66)叶片和根系内的膜脂过氧化程度、细胞膜完整性以及核DNA的影响。结果表明,高温胁迫下:(1)美洲虎3号和TF66的叶片和根系丙二醛(MDA)含量都增加,但两高羊茅品种根系内MDA含量均比叶片内低;(2)两高羊茅品种的叶片和根系内细胞电解质外渗率(EL)均明显增加;(3)同等胁迫水平下,热敏感型TF66的MDA含量和EL水平均高于热耐受型美洲虎3号;(4)两高羊茅品种叶片和根系基因组DNA均没有受到伤害。综合评定的结果,可知较低的MDA含量和EL水平可能是冷季型草坪草抗高温的标志之一,而DNA受伤害状况可能并不适合作为筛选高羊茅耐热性大小的重要指标。
In order to understand the effects of high temperature(35/30°C,day/night,twenty days) on lipid peroxidation, cell membrane thermostability and nuclear DNA of two tall fescue cultivars, Jaguar 3 brand(heat-tolerant) and TF 66(heat-sensitive), a pot experiment was carried out. The results showed that: (1) High temperature stress markedly increased MDA content in leaves and roots of the both cultivars, however, it increased more significantly in leaves than in roots of both cultivars; (2) Levels of EL in leaves and roots of both cultivars increased after high temperature stress; (3) Under the same stress condition, TF 66 retained significantly higher MDA contents and EL levels than Jaguar 3 brand; (4) Very little nuclear DNA damage was found in the two stressed cultivars. It suggests that lower MDA content and EL level may be important indicators associated with tolerance of heat stress in tall fescue turfgrass, while the damage degree of nuclear DNA may be not appropriate index to select heat-tolerant tall fescue turfgrass.
