利用LC-ESI-MS研究丝石竹中的皂苷及其质谱裂解规律

Study on the triterpenoid saponins from the roots of Gypsophila oldhmaniana by HPLC-ESI-MS

为了鉴定丝石竹所含的主要皂苷类成分,采用高效液相-电喷雾离子阱质谱仪(HPLC-ESI-MS)在负离子检测方式下,对丝石竹的皂苷类成分及其裂解途径进行了研究。通过HPLC分析可知主要有六个皂苷成分,通过分析这六个皂苷的碎片信息,发现它具有相似的裂解规律:均是优先裂解C-28酯键所连的糖链,其次裂解C-3所连的糖链,最后生成苷元。这些裂解规律有利于快速发现和鉴定三萜皂苷化合物。

In this paper,six triterpenoid saponins were identified from the radix of Gypsophila oldmaniana by LC-ESI-MS and comparing the retention time with authentic samples. Collision induced dissociation (CID) of the main saponins was performed producing typical degradation reactions which shows the similar fragmentation pathways of six saponins.The cleavage is initiated by a loss of sugar chain of C-28, then loses monosaccharide residue of C-3 sugar chain. Finally aglycones were obtained.The fragmentation pathways were useful for rapid finding and identifying the triterpenoid saponins.
