PGE2, a new regulator of peripheral B cell function
目的:前列腺素E2(PGE2)是一种极其重要的脂类介质,通过EP1-EP4 四种不同受体在多个系统中发挥重要作用。本文主要探索 PGE2 信号通过其 EP2 受体对外周成熟 B 细胞的影响。方法:借助于EP2 缺陷小鼠,检测体内体外 PGE2 对脾中成熟 B 细胞的表型和功能的影响;同时杂交得到 lpr-/-EP2-/- 双基金缺陷小鼠结果,用于检测 PGE2 信号对 B 细胞介导的红斑狼疮样自身免疫疾病的发生发展的影响。结果:EP2 缺陷小鼠的脾脏肿大,原因为脾脏中的 B 细胞数目显著增多。B 细胞表型呈活化状态,并伴有明显的增殖功能增强和凋亡能力减少。 EP2 受体的缺失还能够明显的促进MRL/lpr小鼠的红斑狼疮样病变发病时间的提前和病程加重。 结论:这些数据提示EP2介导的 PGE2 信号对B细胞很可能具有负调控作用,并且 PGE2 信号可能通过其 EP2 受体转导某些信号,在红斑狼疮等自身免疫病发生中的潜在作用。研究结果有助于全面理解B细胞发育和活化的调控机制,拓展对PGE2生理功能的认识,并可能为某些自身免疫病的医学干预提供新的思路。
Aim: Prostaglandin E2 is an important lipid substance, making effect through its EP1-4 receptors. PGE2 play an essential role in many systems. Out study focused on exploring PGE2's influence on peripheral mature B cells through EP2 receptor. Methods: Based on the study of EP2 knockout mice, the phynotype of splenic B cells was screened as well as the proliferation and apoptosis.Moreover, after hybriding EP2-/- mice with MRL/lpr mice, the initiation and accumulation of lupus-like syndromes was carefully examined in order to explore the influence of PGE2 on autoimmune disease mainly induced by B cells. Results: the deficiency of EP2 may make the spleen of knockout mice much larger than wild type mice and their splenic B cells accumulated abnormally.Furthermore, the activation marker such as CD62L and Bcl-2 were damatically changed on EP2-/- B cells. Accordingly, the proliferation of splenic B cells was significantly up-regulted and their apoptosis was down-regulated.After deleting EP2 receptor in MRL/lpr mice, the pathogenisis of lupus-like disease was obviously moved up, accompanying with far more severe of serum anti-self antibodies and kidney tissue destroy. Conclusion: Our data dropped a hint that PGE2 may be a negative regulator for B cells and the PGE2 signal through EP2 receptor would a protential effector in certain autoimmune disease. The result of our research would help to understand the activation and development of B cells better, broaden the cognition of PGE2's physiological effect, and may give a clue for a new medical interfere therapy.
