异种体细胞核移植是在同种体细胞核移植技术的基础上发展起来的,其在保护濒危物种和器官移植上具有重要的意义。本研究的目的是优化水牛-猪异种体细胞核移植(iSCNT)融合/激活参数,筛选出较优融合/激活条件。首先将iSCNT的重构胚在不同的电融合/激活参数下进行融合/激活,再将iSCNT的重构胚分组进行电融合/激活。结果表明电融合/激活中:用200 V/mm,1次脉冲,30μs的直流脉冲为较优融合/激活参数,并且获得iSCNT囊胚。

Interspecies somatic cell nuclear transfer is of great significance for protecting endangered species. The objective of this study was to optimize the parameters of electric fusion/activation used for activating buffalo-pig interspecies somatic cell nuclear transfer (iSCNT) embryos. Firstly, different electro-activation parameters were used for activating iSCNT embryos. The optimized parameters of one 30-μs DC pulse of 200 V/mm were used for activating iSCNT embryos, and this electro-activation method was sufficient for generating buffalo-pig iSCNT.
