RP-HPLC测定麻黄碱手性对应异构体
Separation and Determination of Chiral Ephedrine and Intermediate with PR-HPLC
用RP-HPLC分离测定了生物转化液中的l-麻黄碱、d-伪麻黄碱及前体物质1-苯基-2-甲胺基丙酮(1-phenyl-2-methylamine-acetone,MAK),采用ZORBAX SB-C18柱,UV检测器,以0.02mol/L的V(KH2PO4):V(甲醇):V(醋酸):V(三乙胺)=96:4:0.1:0.13作为流动相,体积流量1mL/min,柱温35℃,检测波长210nm,MAK、l-麻黄碱、d-伪麻黄碱的检出限分别为0.05,0.2,0.1mg/L,分别测定了两株菌生物转化液中的MAK、l-麻黄碱、d-伪麻黄碱的质量浓度,回收率为97.2%~99.3%。
sensitive and reliable high performance liquid chromatographic method(HPLC)has been developed for the simultaneous determination of l-ephedrine, d-pseudoephedrine and a intermediate 1-phenyl-2-methylamine (MAK) in bioconversion. The HPLC assay was performed on a reversed phase C18 column (ZORBAX SB C18 5 μm 4.6 mm×250 mm) by using methanol-0.2mol/L KH2PO4-acetic acid-triethyl amine (4:96:0.1:0.13,V/V) as mobile phase for analysis. Regression equations revealed the liner relationships (correlation coefficients: 0.9999) between the peak area of each constituent (ephedrine, pseudoephedrine and MAK) and its concentration, The detection limits(S/N=3) for MAK, ephedrine and pseudoephedrine were 0.05mg/L,0.2mg/L and 0.1mg/L, respectively. The bioconversions of the two strains were determined and the recoveries ranged between 97.2%-99.3%.
