目的:观察硫氧还蛋白还原酶(TrxR)在石英诱导的大鼠肺纤维化中的表达,以及丹参脂质体干预对TrxR表达的影响。方法:按随机数字表法将90只Wistar大鼠分为正常生理盐水对照组(对照组)、石英染尘模型组(模型组)、丹参脂质体干预组(药物组),制备大鼠肺纤维化模型,分别于染尘后的第7、15、30天宰杀大鼠并提取肺组织,应用DCFH-DA探针法检测肺组织中ROS,硫代巴比妥酸法检测MDA含量,Western Blot法分析每组不同时间点肺组织中TrxR蛋白表达。结果:石英染尘可加重大鼠肺部的氧化应激,7天模型组中ROS、MDA含量明显高于对照组(P<0.05),TrxR蛋白表达明显低于对照组(P <0.05),而15天出现代偿性表达上调(P<0.05);与模型组比较,药物组ROS、MDA含量在7天和15天较模型组均有降低(P<0.05),并纠正在氧化应激早期引起的TrxR表达下调。结论:丹参脂质体对大鼠肺纤维化有一定保护作用,其机制可能与调节氧化抗氧化平衡、抑制脂质过氧化发生有关。

his study aimed to investigate the possible mechanism of Salvia miltiorrhizaliposome(SalmL) to alleviate the pulmonary fibrosis induced by silica in vivo, in which thioredoxin reductase (TrxR)may play the role as an antioxidant enzyme. Rats were randomly divided into saline control group, silica group and SalmL preventive treatment group for 7, 15, 30, or 60 days consecutive experiment. Our results demonstrated silica could significantly decrease the expression of TrxR, resulting in the accumulation of reactive oxygen species (ROS) and malondialdehyde (MDA) during the early period. However, in SalmL group, the expression of TR was much higher than the silica treated group and the ROS accumulation was less. In conclusion, SalmL could restore TrxR expression decreased by silica to exert its inherent antioxidative function and alleviate the processing of pulmonary fibrosis induced by silica.
