瑞士乳杆菌MB2-1胞外多糖发酵条件的优化
Optimization of Fermentation Conditions for Production of Exopolysaccharides by Lactobacillus helveticus MB2-1
对一株分离自新疆赛里木酸奶中的瑞士乳杆菌MB2-1(Lactobacillus helveticus MB2-1)胞外多糖(EPS)的发酵条件进行优化研究。利用单因素实验确定发酵温度、乳糖添加量、氮源种类以及添加量、无机盐种类以及添加量对其EPS产量的影响。采用Box-Benhnken法对其中的3因素3水平进行响应面分析以确定其最优工艺条件。结果表明:L. helveticus MB2-1 EPS提取的最佳条件为发酵温度37℃、Mg2+添加量为质量浓度0.2%、大豆蛋白胨添加量为质量浓度2%,在此工艺条件下,L. helveticus MB2-1 EPS的产量达到801.2mg/L。结论:应用Box-Benhnken设计和响应面法进行L. helveticus MB2-1 EPS提取条件的优化是可行的。
he fermentation conditions for the yield of exopolysaccharides (EPS) by Lactobacillus helveticus strain of MB2-1 isolated from Sayram traditional yogurt from Xinjiang were optimized. The effects of culture temperature, lactose concentrations, nitrogen sources and concentrations, mineral sources and concentrations on production of EPS were studied using the method of single factor analysis. On the 3 levels of response surface analysis (RSM) were carried to determine the optimal process conditions by using Box-Benhnken method. Results showed that the optimum extraction conditions were culture temperature of 37℃, Mg2+ concentrations of 0.2 %, soybean peptone concentrations of 2 %. Under these conditions, the yield of EPS by L. helveticus MB2-1 was obtained to 801.2 mg/L. Conclusion: It is feasible to apply Box-Benhnken design and RSM to optimize the extraction conditions for the production of EPS by L. helveticus MB2-1.
