地塞米松加重OPG缺乏引发的骨质疏松
OPG increase dexamethasone induced osteoporosis
【目的】研究在糖皮质激素引发的骨质疏松中OPG的作用。【方法】向OPG敲除小鼠和野生型小鼠腹腔注射地塞米松,组织学切片行HE染色和TRAP染色,观察股骨骨小梁和骨皮质区域骨量和破骨细胞的变化。【结果】地塞米松导致小鼠股骨骨小梁和骨皮质的骨量下降。在OPG缺失的条件下,地塞米松引发的骨量下降更加显著,破骨细胞数量明显增加。【结论】OPG在地塞米松引发的小鼠骨质疏松中,减少骨量的丢失,抑制破骨细胞的数量。糖皮质激素在引发骨质疏松的过程中,可能和OPG的表达抑制具有协同作用。
【Objective】 To study the effect of OPG on osteoclast in glucocorticoid-induced osteoporosis. 【Methods】 Dexamethasone was injected intraperitoneally into OPG knockout mice and wild type mice. HE staining and TRAP staining were used to observe the changes of bone mass and osteoclasts in femoral trabeculae and cortical area. 【Results】 Dexamethasone resulted in decreased bone mass of trabecular and cortical bone in mice. In the absence of OPG conditions, dexamethasone caused bone mass decreased more significantly, the number of osteoclasts increased significantly. 【Conclusion】 OPG reduces the loss of bone mass and inhibits the number of osteoclasts in dexamethasone-induced osteoporosis in mice. OPG suppression may contribute to glucocorticoids induced osteoporosis.
