目的:急性中风是临床上得常见病,具有较高的致残率和致死率。本研究旨在阐述胆酸类成分与栀子苷在神经保护方面的相互作用效果及机制,以调整JZQKL配方,筛选最佳组合。方法:本文研究了栀子苷与胆酸类成分组合在OGD-Rep模型下的SH-SY5Y细胞的影响,通过CCK-8测定细胞活性,应用流式细胞术检测细胞凋亡情况,并检测了细胞上清中的LDH。结果:低剂量0.5μg/mL HDCA与栀子苷合用后,基本未影响细胞活性,而5、10μg/mL与栀子苷合用后则表现细胞活性下降趋势;CA与HDCA的表现类似。而TUDCA 0.5、5μg/mL与栀子苷合用时细胞活力较高,GT组合较单纯Gen略高一点,差异无统计学意义。当TUDCA剂量增大时,达到50~100μg/mL时,会减弱Gen对细胞的保护作用,增加对细胞的损伤。结论:栀子苷与TUDCA合用能够发挥最佳的神经保护作用,栀子苷与TUDCA组合初步订为20:1。

Objective: acute stroke is the clinical common disease, has the high morbidity and mortality. This study aims to expound bile acids composition and the interaction of geniposide in neural protection effect and mechanism, to adjust JZQKL formula, screening the best combination. Methods: this paper studies the gardenoside and bile acid composition in OGD - Rep model under the influence of SH SY5Y cells, through the determination of CCK 8 cell activity, application of flow cytometry to detect cell apoptosis, and tested the cell supernatant of LDH. Results: low doses of 0.5 mu g/mL HDCA and geniposide pooling, basic did not affect cell activity, and 5, 10 mu g/mL and geniposide share after the cell activity decline; CA is similar to HDCA performance. TUDCA, 5 mu 0.5 g/mL and geniposide share when high cell vitality, GT combination relatively simple Gen slightly higher, there was no statistically significant difference. When TUDCA dose increased, up to 50 ~ 100 mu g/mL, weaken Gen protection of cells, increase the damage in cells. Conclusion: the gardenoside and TUDCA share can play the best nerve protective effect, gardenoside and TUDCA combination preliminary order for those days.
