苦瓜BAG基因组织特异性表达研究

实时荧光定量RT-PCR广泛应用于定量检测mRNA表达水平,具有准确、灵敏等特点。本文通过实时荧光定量RT-PCR分析了苦瓜MADS-box基因BAG在发育各个器官和组织的表达情况,结果表明:BAG基因仅在生殖器官中表达,花瓣中没有表达,心皮中表达水平远远高于其它组织,为雄蕊的77倍之多;在幼果和花萼中也检测到有低丰度的表达。分析表明BAG基因直接参与苦瓜花和果实发育,尤其对心皮的发育起着关键的作用。
In the present study, we developed a new, sensitive, reproducible and reliable real-time quantitative RT-PCR assay for measuring the BAG mRNA expression level of Momordica charantia L. The result showed that the expression of BAG was only present in the reproductive organs. High mRNA levels were present in carpel and moderate level in stamen, calyx and young fruit. All these data demonstrated BAG gene participates in development of flower and fruit, especially having a key role in carpel development.
