NA损伤与很多人类重要的疾病相关,评估DNA损伤的程度对于研究疾病的发病机制和评估药物以及营养物质对于DNA损伤修复的能力具有重要的意义。本研究运用原子力显微镜(AFM)观测正常DNA分子、损伤DNA分子,通过对DNA分子名义覆盖面积的分析,得到了这两种DNA分子大小的明显差异。通过对116个不同的DNA分子长轴和短轴的分析,得出了正常DNA分子名义覆盖面积明显大于损伤DNA名义覆盖面积的结论。这一点与DNA损伤后分子量变小的事实相符。因此可以利用名义覆盖面积这一变量,通过AFM评估DNA损伤程度,进而将其应用于DNA损伤程度和DNA修复程度的评估。

DNA damage is an important factor of many diseases. It is significant to develop the new technique for observation and evaluation of the degree of DNA damage for studying the pathogenesis of diseases and assessment repair ability of drugs and nutrients. The natural and damage DNA molecules were observed by atomic force microscopy (AFM) in present study. One hundred and sixteen AFM images of different DNA molecules were analyzed. They showed significant difference in nominal covering area, which is consistent with the phenomenon of DNA molecular weight degradation after oxidative damage. It is suggested that AFM is a direct method for observation of DNA damage, which can be used to evaluate DNA damage or repair.
