【目的】:为了在动物水平进行TLR2受体功能研究,开展了针对小鼠TLR2胞外区 (TLR2-ECD)单克隆抗体的制备工作。【方法】:利用RT-PCR 方法扩增出小鼠TLR2胞外区基因并将其克隆到原核表达载体pGEX-4T-1 上,构建重组表达质粒pGEX-TLR2-ECD,并将其转化到大肠杆菌BL-21 中,在诱导剂IPTG 的诱导下获得高效表达。重组融合蛋白TLR2-ECD-GST的分子量约为90KDa。用纯化的重组蛋白TLR2-ECD-GST免疫Bal b/c 小鼠,采用杂交瘤技术制备抗TLR2-ECD蛋白的单克隆抗体。通过细胞融合技术获得可分泌抗体的杂交瘤细胞株,经间接酶联免疫吸附试验阴性筛选和阳性筛选步骤,获得能够分泌特异性抗TLR2-ECD抗体的杂交瘤细胞株。【结果】:获得分泌TLR2-ECD单克隆抗体的细胞株。【结论】:成功制备分泌TLR2-ECD单克隆抗体的细胞株,并鉴定该细胞株分泌的单克隆抗体特异性识别TLR2胞外区,与融合标签蛋白GST没有交叉反应。

【Objectives】: In order to provide tools for studying the function of TLR2 receptor in tumor metastases, the monoclonal antibodies against the extracellular domain of mouse TLR2 were prepared. 【Methods】: The extracellular domain of mouse TLR2 was cloned into the prokaryotic expression vector pGEX-4T-1. Then the E. coli strain BL21 was transformed with the recombinant plasmid pGEX-TLR2-ECD and the fusion protein TLR2-ECD-GST was highly expressed by IPTG induction. Bal b/c mice were immuned with purified recombinant protein TLR2-ECD-GST, and the hybridoma was prepared to produce monoclonal antibodies against TLR2-ECD. Following negative screening and positive selection steps by indirect ELISA, hybridoma cell strains with the ability to secrete specific antibodies against TLR2-ECD were identified. 【Results】: Hybridoma cell strains secreting monoclonal antibody against TLR2-ECD were prepared. 【Conclusions】: Hybridoma cell strains secreting monoclonal antibody specifically recognized by TLR2 extra cellular domain were established. No cross-reactivity of the antibodies with the GST tagged protein was observed.
