目的 确定黄芩苷脂质体的最佳制备工艺。方法 以逆相蒸发法制备黄芩苷脂质体,以包封率为评价指标,采用单因素考察和正交设计试验进行系统研究,考察了水相与有机相的比例、药物浓度、超声时间、水浴温度、旋转蒸发仪转速、大豆卵磷脂用量及类脂比等因素对脂质体包封效果的影响,从而确定黄芩苷脂质体的最佳制备工艺。结果 确定黄芩苷脂质体最佳制备工艺条件为:水相与有机相比例为1:4、黄芩苷投入量(药物浓度)为5mg/ml、磷脂用量为200g、卵磷脂与胆固醇比例为1:6,制得的脂质体平均粒径为178nm,包封率为50.3%。结论 逆相蒸发法制备的黄芩苷脂质体粒径小,包封率高,条件易掌握,可作为黄芩苷脂质体的常规制备方法。

Objective:To study the preparing technique of baicalin liposomes Method: Bacalin liposomes were prepared by reverse-phase evaporation, A reasonable criterion of accurate evaluation was established by which such factors as the ratio of organic phase to aqueous phase ,rotation speed, waterbath temperature,hypersonic time, and the concentration of reactants were examined and the orthographic design was used and the best preparation technology is determined. Results: The optimized preparation conditions for baicalin phytosomes were obtained as follows: the ratio of aqueous phase to organic phase was 1 to 4. the dosage of phospholipid was 200mg.The ratio of phospholipid to cholesterin was 1 to 6. Baicalin concentration was 5 mg/ml. the mean diameter of liposomes was 178nm, envelopment percentage was 50.3%. Results: the optimized preparation is feasible and stable.
