目的:探讨α-1,6岩藻糖基转移酶(α1,6-fucosyltransferase 8,FUT8)对嘌呤霉素氨基核苷(puromycin aminonucleoside,PAN)致小鼠足细胞损伤的影响。方法: 体外培养永生化小鼠足细胞系(mouse podocyte clone 5,MPC5),37 C条件下培养诱导其分化后,利用PAN刺激建立足细胞损伤模型,利用RNAi技术敲低足细胞FUT8的表达,荧光实时定量PCR方法测定各组细胞FUT8mRNA的表达,免疫荧光方法测定各组细胞表面核心岩藻糖链和足细胞特异标志物Nephrin的表达,免疫印迹方法测定各组FUT8、Nephrin的蛋白表达,流式细胞术测定各组细胞凋亡。结果:1.免疫荧光显示MPC5细胞上存在中等量核心岩藻糖链表达。2. FUT8siRNA转染后PAN加FUT8siRNA组与PAN组相比,从培养皿底脱落细胞数明显减少,MPC5细胞表面核心岩藻糖链和FUT8的表达降低, Nephrin蛋白表达量增多,细胞凋亡数减少。结论:1.核心岩藻糖基化修饰参与了嘌呤霉素氨基核苷诱导的小鼠足细胞凋亡和结构蛋白的改变。2.应用RNA干扰技术沉默FUT8基因,阻断足细胞的核心岩藻糖基化修饰,能在一定程度上抑制嘌呤霉素氨基核苷诱导的小鼠足细胞损伤。

Objective:To investigate the effects of α1,6-fucosyltransferase 8(FUT8) on murine podocyte injury caused by puromycin aminonucleoside(PAN). Methods: A conditionally immortalized mouse podocyte clonal cell line (mouse podocyte clone 5, MPC5) was cultured in medium containing mouse γ-interferon at 33 C in vitro,FUT8siRNA suppress the expression of FUT8 in podocytes. the expression of FUT8mRNA was detected by RT-PCR, the expression of core fucose and podocyte marker-Nephrin was detected by immunofluorescent analysis, the expression of FUT8 and Nephrin was detected by western-blotting, and the apoptosis was examined by flow cytometry. Results:1. It was confirmed by immunofluorescence that core fucose existed in MPC5. 2. immunofluorescent analysis showed that the expression of core fucose was significantly increased after PAN incubation, while Nephrin was decreased; western- blotting showed that the expression of FUT8 was significantly increased, while Nephrin was decreased after PAN incubation; the apoptosis of cells was increased after PAN incubation. Preincubation with FUT8siRNA could inhibit cells detachment from the bottom of the culture dish;the expression of core fucose and FUT8 was decreased, while Nephrin was increased; the apoptosis of cells was decreased.Conclusion: 1. The core fucosylation involved in the apoptosis and the change of structural protein in podocyte injury caused by puromycin aminonucleoside.2. Utilizing RNAi technology to silence the expression of FUT8 gene , blocking of the core fucosylation, led to inhibition of podocyte injury induced by puromycin aminonucleoside.
