高山被孢霉常温高产EPA的研究
Study on eicosapentaenoic acid production of Mortierella alpina at room temperature
二十碳五烯酸(EPA; C20:5; ω-3)是一种ω-3系列长链多不饱和脂肪酸,具有抗凝血、防心血管病、降血脂等特殊生理功能,在保健品、食品和饲料行业均有较多的应用。产油微生物高山被孢霉是花生四烯酸(AA; C20:4; ω-6)生产的工业化菌株,其AA产量约占总脂肪酸的50%,但EPA的合成量却很低。ω-3脂肪酸脱饱和酶具有催化AA生成EPA的作用,是ω-3长链多不饱和脂肪酸合成途径的关键酶。本文为了提高高山被孢霉的EPA产量,将来源于瓜果腐霉偏好催化AA的ω-3脂肪酸脱饱和酶PaD17在高山被孢霉体内过表达,共得到六株重组菌,其中重组菌株CCFM 695的EPA产量在常温下高达600 mg/L以上。通过单因素和正交实验对其发酵培养基的碳氮源进行优化,结果表明以葡萄糖和豆粕作组合碳氮源条件下,EPA产量高达1.7 g/L,比原始发酵条件增产约3倍。
Eicosapentaenoic acid is an omega-3 long chain polyunsaturated fatty acid with special physiological functions, such as anticoagulant, anti-cardiovascular disease and hypolipidemic. It has many applications on health care products, food and feed industry. Oleaginous fungus Mortierella alpina is an industrial strain of arachidonic acid production, but the amount of EPA is very low.Omega-3 fatty acid desaturase is a key enzyme in the synthesis of omega-3 long chain polyunsaturated fatty acids and can catalyzed AA to generate EPA. To enhance EPA production, we heterologous expressed the Pythium aphanidermatumω-3 fatty acid desaturase PaD17in Mortierella alpina and obtained six recombinantstrains.The EPA yield of the recombinant strain CCFM695 was up to 600 mg/Lat room temperature.The effects of different carbon and nitrogen sources on the accumulation of EPA were studied by single factor and orthogonal experiments. Results showed that the yield of EPA was up to 1.7 g/L under the condition of glucose and defattedsoybean meal, about 3 times more than the original fermentation conditions.
