手机辐射对鸡胚心脏发育的影响
Effects of mobile phone radiation on heart development in chick embryo
目的 探讨手机(MP)辐射对鸡胚心脏发育过程中心肌显微、超微结构的改变和心脏内血细胞DNA的损伤,以及心肌组织内Chk1表达的变化及其机制。方法 海兰白鸡受精卵随机分为对照组和辐射组,辐射组鸡胚均于孵化第2至21d每天接受3h整体手机辐射(900/1,900 MHz; SAR≈1.07W/kg),应用光镜和电镜、彗星电泳及Western blot 技术来研究MP辐射对鸡胚心脏的影响。结果 鸡胚发育早期辐射组心脏无明显病理学改变,胚胎发育中期,辐射组心肌组织内脂滴明显增多,随着辐射剂量累积,至胚胎发育晚期(15-21d),辐射组部分心肌纤维出现断裂、轻微溶解或脂肪样变化,心肌细胞核肿胀,核周间隙扩张,线粒体空泡化等;辐射组鸡胚发育到第15d时,心脏血细胞拖尾面积显著高于对照组(P<0.05);在鸡胚发育的早、中期,对照和辐射组心脏中Chk1的表达量均较高,但无统计学差异,至21d时Chk1在对照组心脏中表达消失,而实验组中却有一定量的表达。结论 一定剂量的MP辐射能在非热效应下造成心肌细胞显微、超微结构及细胞DNA损伤,并且使Chk1基因过表达,而参与了DNA修复或细胞凋亡的发生过程。
o investigate the effects of Mobile Phone Radiation (MP) on microscopic structure, ultrastructure and Chk1 expression of heart development in chick embryo, DNA damages in blood cells. [Methods] The chick embryos were randomly and evenly divided into control group and exposure group. Embryos in exposure group were irradiated by MP (900/1,900 MHz; SAR≈1.07W/kg) 3h per day from day 2 to day 21 of the incubation of the embryo. Light microscope, electron microscope techniques, single cell gel electrophoresis and western blot were used to study the effects.[Results] Compared to the control group, exposure group had no obvious lesion changes in early embryo development, on the 10th and 13th day , the distribution of lipid droplet in exposure group increased significantly. From day 15 to day 21, the MP radiation caused dispersed disruption of myofilament, focal necrosis of the atrioventricular valve, vacuolization and deformation of mitochondrium in exposure group were evidenced. On the 15th day, the tail area in blood cells in chick embryo heart in the exposure group was higher than that in the control group (p<0.05). Chk1 expression was much higher in both control and exposure group and no obvious difference in early and middle embryo development. But on the 21st day Chk1 expression was only in exposure group. [Conclusions] Mobile Phone Radiation may cause microscopic structure and ultrastructure alterations of myocardial cells, and induce DNA damages. The expression of Chk1 was increased significantly and played an important role in the DNA repairing and cell apoptosis process.
