ODD调节HSV-TK/GCV对乏氧环境宫颈癌Hela细胞特异杀伤作用的研究
ODDD regulated herpes simplex virus-thymidine kinase/gancyclovir system killing effect on Hela cell line under hypoxia
目的 构建一种由氧依赖降解结构域(ODDD)调控的单纯疱疹病毒胸苷激酶(HSV-TK)基因表达的真核表达载体,观察ODDD能否使HSV—TK/GCV系统对乏氧环境中的宫颈癌Hela细胞有特异杀伤作用。方法 RT-PCR从Hela细胞中钓取ODDD 片段,将其克隆入荧光报告载体pEGFP-C1,构建pEGFP-ODDD;扩增HSV—TK基因,将其克隆入pEGFP-ODDD和pEGFP-C1的下游,构建pEGFP-ODDD-TK及pEGFP-TK。将pEGFP-ODDD转染入Hela细胞,流式细胞术检测在乏氧(O2浓度为1%)和常氧(O2浓度为21%)条件下细胞中EGFP表达强度。以GCV处理转染pEGFP-ODDD-TK或pEGFP-TK的Hela细胞,MTT法检测GCV对培养细胞的杀伤效果。结果 经酶切分析、PCR和DNA序列测定证实,构建了含有ODDD序列调控的HSV-TK真核表达载体;含有ODDD片段荧光报告载体的Hela细胞在常氧浓度下EGFP的表达量为11.96%低于乏氧条件下的39.92%;在乏氧环境下,转染pEGFP-ODDD-TK的Hela细胞对GCV的敏感性高于常氧含量组(P<0.01),而转染pEGFP-TK的Hela细胞在常氧和乏氧条件下对GCV的敏感度基本相同。结论 构建了含有ODDD序列调控的HSV-TK真核表达载体;常氧环境下,ODDD使报告基因EGFP表达降低。ODDD能特异发挥HSV-TK/GCV系统对乏氧环境Hela细胞的杀伤作用。
0bjective To observe the specific killing effects of a eukaryotic expression vector expressing herpes simplex virus thymidine kinase regulated by ODDD(oxygen dependent degradation domain)on Hela cell under hypoxia which makes it possible to further investigate their potency of antitumor potency under hypoxia. Methods The encoding sequence for ODDD amplified with RT-PCR and inserted into pEGFP-C1;The HSV—TK was amplified by PCR and cloned into the multiple clone site of pEGFP-C1 and pEGFP-ODDD to obtain pEGFP-TK and pEGFP-ODDD-TK recombinant plasmid.Transient transfected cells with pEGFP-ODDD were exposed to hypoxia 24 h after transfection. Relative EGFP fluorescence was measured by a flow cytometer under hypoxia and normoxia . The sensitivity of Hela cell line transfected with pEGFP-ODDD-TK and pEGFP-TK recombinant plasmid to the drug GCV was determined with the method of MTT after treating with GCV for five days.Results The integrity reporter vectors were verified by restriction enzyme digestion. the EGFP expression levels of pEGFP-ODDD in normoxia in Hela cells were 11.96% and were lower than those of hypoxia sample39.92%. Comparison of the sensitivity of cells to GCV showed that the cells in pEGFP-ODDD-TK groups under hypoxia were much more sensitive to GCV than the cells under normoxic condition(P<0.01),the higher the GCV concentration,the greater the difference .Conclusion ODDD could ruduce expression of EGFP in Hela cells under normoxic condition. ODDD could specify the kiling effect of HSV-TK /GCV system on Hela cell line under hypoxic condition.
