以水稻抗坏血酸过氧化物酶OsAPX2为转化对象,采用农杆菌介导子叶节转化技术将其转入大豆品种冀豆12,转基因T0和T1植株均可扩增出1268 bp目的条带,测序分析证实与转化基因序列一致,说明OsAPX2已成功整合于大豆基因组;RT-PCR结果显示,OsAPX2能够在转基因植株中正常表达,qPCR分析发现5个转基因株系以L33、L34的OsAPX2表达量较高;干旱处理条件下,L33、L34转基因株系的抗旱性明显优于野生型对照,OsAPX2具有提高转基因大豆抗旱性的功能。

In this study, putative OsAPX2-over-expression soybean lines were obtained by Agrobacterium-mediated cotyledonary-node method. Subsequently, PCR and DNA sequencing were performed to detect the transgenic lines. The results of PCR amplification and DNA-sequencing in T0 and T1 plants showed that 1268 bp bands were clearly electrophoresed on the gel, and the sequence of the bands were consistent with the original sequence of OsAPX2, which demonstrated that OsAPX2 were successfully integrated into the genome of soybean. The results of RT-PCR showed that OsAPX2 were stably expressed in the transgenic soybean plants, and the transcript accumulations of L33 and L34 transgenic lines were significantly increased than the others. Drought resistance determination showed that these two transgenic lines (L33, L34) had stronger capability to tolerate drought stress, compared with wild-type, which suggested that over-expression of OsAPX2 might enhance soybean drought-resistance.
