目的:本研究拟考察D-半乳糖对小鼠学习记忆功能影响,并初步探讨其可能的作用机制。方法:将3月龄雄性ICR小鼠随机分为正常对照、D-半乳糖组。D-半乳糖组小鼠颈背部皮下注射无菌D-半乳糖液(1000 mg/kg),对照组注射等容量生理盐水。连续注射10周后,采用Morris水迷宫实验考察D-半乳糖对小鼠学习记忆能力的影响;采用Western Blotting方法考察D-半乳糖对小鼠海马mTOR蛋白的影响。结果:与正常组比较,D-半乳糖能明显延长小鼠水迷宫潜伏期。Western Blotting结果显示D-半乳糖能降低小鼠海马组织中P-mTOR蛋白含量。结论:D-半乳糖能损伤小鼠学习记忆功能,这可能与抑制海马mTOR蛋白表达有关。

Objective: This study intends to investigate the effects D-galactose on learning and memory in mice, and to explore its possible mechanism. Methods: 3-month-old healthy male ICR mice were randomly divided into normal control and D-galactose groups. Mice in D-galactose group were received daily subcutaneous injection of D-galactose at a dose of 1000 mg/kg and the normal group were received the same volume saline. After 10 weeks of injection, effect of D-galactose on the function of learning and memory was investigated by Morris water maze. The effect of D-galactose on mTOR protein in the hippocampus of mice was explored by Western Blotting method. Results:D-galactose can significantly prolong the incubation period in water maze and reduce P-mTOR protein content in mice hippocampus compared with normal group. Conclusion:D-galactose can damage the function of learning and memory in mice, it may be associated with inhibiting mTOR protein expression in hippocampus.
