目的 为探讨阿托伐他汀控制自发性高血压大鼠(SHR)高血压状态的机制,研究阿托伐他汀对SHR的血管平滑肌细胞调亡和p27蛋白表达的影响。方法 选用8周龄SHR12只,随机分为阿托伐他汀治疗组(ATV组, n=6)和SHR组(n=6),并以同周龄WKY(n=6)作为对照。ATV组给以阿托伐他汀(50mg.kg-1.d-1)灌胃,SHR组和WKY组给于等量蒸馏水灌胃。10周后观察三组大鼠血压、血清总胆固醇(TC)、总甘油三酯(TG)的变化,以及TUNEL法检测主动脉平滑肌细胞(ASMC)凋亡率,测定动脉ASMC p27蛋白表达。结果 阿托伐他汀给药10周后,ATV组动脉收缩压显著低于SHR组(134.17±3.60 vs. 173.33±3.78 mmHg, P<0.01);ATV组血清TC和TG浓度较SHR组均显著降低(P<0.01, P<0.01); ATV组ASMC凋亡率较SHR组显著增高(16.94±3.08 vs. 9.01±2.36%, P<0.01);与WKY大鼠比较,ATV组ASMC p27蛋白表达阳性率显著增高(24.25±4.41 vs. 33.02±5.01%, P<0.05),而SHR组该指标明显降低(24.25±4.41 vs. 16.08±7.09%, P<0.01)。结论 阿托伐他汀控制SHR血压增高,其机制可能与增高ASMC凋亡率和p27蛋白表达阳性率有关。

IM The aim of this study was to evaluate the effects of atorvastatin on aortic smooth muscle cells (ASMC) apoptosis and p27 expression in spontaneously hypertensive rats (SHR). METHODS 12 eight-week-old SHR were randomized into atorvastatin group (ATV, n=6) and SHR group (n=6). 6 age-matched normotensive Wistar-Kyoto rats (WKY) were as controls. 50mg.kg-1.d-1 of atorvastatin was administered to ATV by gavage for 10 weeks. Serum cholesterol and triglycerides were measured, and systolic blood pressure of caudal artery was examined. ASMC apoptosis rate was detected by TUNEL technique, and positive expression rate of p27 in ASMC was analyzed. RESULTS After 10 weeks, systolic blood pressure in ATV was significantly lower than that in SHR (134.17±3.60 vs. 173.33±3.78 mmHg, P<0.01). Compared with SHR, serum cholesterol and triglycerides were significantly lower (P<0.01, P<0.01) in ATV. ASMC apoptosis rate was higher in ATV than that in SHR (16.94±3.08 vs. 9.01±2.36%, P<0.01). Compared with WKY, positive expression rate of p27 in ASMC from ATV was higher (24.25±4.41 vs. 33.02±5.01%, P<0.05), whereas that was lower in SHR (24.25±4.41 vs. 16.08±7.09%, P<0.01). CONCLUSION Atorvastatin may up-regulate p27 expression in ASMC and facilitate ASMC apoptosis which may effectively reduce blood pressure in SHR.
