利用线虫研究多不饱和脂肪酸对肠道菌群的调节作用

Polyunsaturated fatty acids impact intestinal microbes based on the model of Caenorhabditis elegans

肠道微生物对人体健康起到重要的调节作用,而通过饮食调节可以使肠道菌群向对人体有益的状态发展。线虫具有生命周期短、以细菌为食、易培养等特点,已应用于益生菌和致病菌等的研究中。本研究主要采用二代测序技术对菌群宏基因组进行测定,分析了引入线虫体内的菌群组成及分布,验证了利用线虫研究菌群的可行性;比较两种食物组分n-3PUFAs和n-6PUFAs对肠道菌群丰度、多样性及组成产生的影响。并以线虫寿命为指标进一步测定受到n-3PUFAs和n-6PUFAs调节的肠道菌群对机体生命活动的影响。结果表明线虫体内能够引入小鼠粪便菌中的优势菌群,两种多不饱和脂肪酸对肠道菌群组成和丰度产生不同影响,且受n-3PUFAs调节的菌群与n-6PUFAs调节的菌群相比能够延长线虫寿命。本研究表明线虫具有快速有效筛选能够调节肠道菌群的食物组分的潜力,并可用于观察受调节菌群对机体生命活动的影响,为食物组分对肠道菌群作用的研究提供了新模型及新思路。

Intestinal microbes are important to maintenance human health. People can obtain healthier intestinal microbes by modulating daily diets. Caenorhabditis elegans (C.elegans) is charaterized with short lifespan, easy to culture and feeding on bacteria. It has been used to study probiotics and pathengenics. In the paper, genome analysis was performed using the second generation sequencing to analyze the composition of microflora introduced to C.elegans and the differences of the diversity and composition of intestinal microbes caused by n-3PUFAs and n-6PUFAs. Furthermore, lifespan of C.elegans was used as an indicator to detect the influences of intestinal microbes modulated by n-3 or n-6PUFAs has on the host's life-history traits. The result showed that major bacterial groups of intestinal microbes from mice could be introduced into C.elegans. There were differences in the diversity and composition in microflora between n-3PUFAs and n-6PUFAs groups. C.elegans feeding on microflora from mice with n-3PUFAs-rich diets tended to live longer than the others. In the paper, C.elegans provided a fantastic opportunity to study the interactions between bacteria and the hosts. This will help to establish a rapid and efficient screening model for food components related to the modulation of intestinal microbes as well as a physiological model to explore potential impacts of the intestinal microbes under modulation.
