哺乳妇女外周血中TSHR、PKA及NIS mRNA表达的相关分析

目的:检测哺乳妇女外周血中TSH-TSHR-cAMP-PKA信号通路相关基因mRNA的表达变化,探讨TSH-TSHR-cAMP-PKA信号通路在哺乳妇女甲状腺疾病中的作用。方法:在山西省适碘和高碘地区采集99例哺乳妇女外周血,其中甲状腺功能异常的51例,甲状腺功能正常的健康对照48例。用实时荧光定量PCR法检测TSHR,PKA及NIS mRNA的表达水平。结果:病例组TSHR、PKA和NIS的表达量分别为3.57±6.68、1.59±2.43、2.77±4.21,均高于对照组相应的结果1.77±2.89、1.16±1.31、1.69±4.89,但差异均无统计学意义(P>0.05)。 TSHR与NIS在病例组和对照组中均为显著正相关(r(病)=0.862,P<0.01;r(对)=0.459,P<0.01);PKA与NIS病例组之间为显著正相关(r(病)=0.452,P<0.01),对照组之间未见明显相关性;TSHR与PKA未见明显相关性。高碘地区TSHR mRNA的表达量明显低于适碘地区,差异有统计学意义(P<0.05);高碘地区PKA和NIS mRNA表达量均低于适碘地区,差异无统计学意义(P>0.05)。结论:哺乳期妇女可能通过TSH-TSHR-cAMP-PKA信号通路调控机体的摄碘水平来达到保护自身及子代目的。
Objective:To explore the role of the TSH-TSHR-cAMP-PKA signaling pathway in thyroid disease of lactating women,we detected the expression changes of the TSH-TSHR-cAMP-PKA signaling pathway related genes mRNA in peripheral blood .Methods:A total of 101 samples of peripheral blood of breast-feeding women were collected in Shanxi province's sufficient iodine and high iodine areas.There are 51 cases of thyroid dysfunction and 48 healthy controls.The mRNA expression of TSHR,PKA and NIS were detected by real-time quantitative PCR.Results:The mRNA expression of TSHR,PKA and NIS in case group (3.57±6.68,.59±2.43,.77±4.21)were higher than that of control group(1.77±2.89,1.16±1.31,1.69±4.89),but the differences were not statistically significant(P>0.05).The TSHR and the NIS in the case group and control group were significantly positively correlated((r(d)=0.862,P<0.01;r(c)=0.459,P<0.01);The PKA and the NIS in the case group was a significant positive correlation (r(d)=0.452,P<0.01),but in the control group there was no significant correlation(P>0.05);The TSHR had no significant correlation with the PKA.The TSHR mRNA expression in waterborne high iodine areas was significantly lower than the sufficient iodine areas,the difference was statistically significant(P<0.05);the PKA and NIS mRNA expression in high iodine area were lower than the sufficient iodine regions, the difference was not statistically significant (P> 0.05).Conclusion:Lactating women may be to protect themselves and their babies through the TSH-TSHR-cAMP-PKA signaling pathway regulating the iodine levels.
