苦马豆素抗肿瘤作用的研究进展

苦马豆素是一种吲哚里西啶生物碱,最早是从灰苦马豆中分离得到,从发现至今已有近30年的历史。它是一种极强的α-甘露糖苷酶竞争性抑制剂,能抑制肿瘤细胞表面糖蛋白的表达,从而抑制肿瘤细胞的生长与转移,同时还能刺激机体的免疫系统,提高机体的免疫机能,促进免疫细胞的增生,增强杀灭肿瘤细胞的能力。由于苦马豆素具有很好的抗肿瘤活性和免疫增强作用,已引起国内学者的广泛关注,并作为抗肿瘤药筛选的后备药。目前,国外苦马豆素抗肿瘤药的研发已进入Ⅲ期临床试验阶段,而国内尚处于起步阶段。本文主要从直接和间接两方面介绍了苦马豆素对肿瘤细胞抑制作用的研究进展,并对苦马豆素在临床治疗中取得的成果进行综述。

he indolizidine alkaloid swainsonine was initially isolated from Swainsona canescens, which was about thirty years up to now. Swainsonine, an highly specific alpha-mannosidase competitive inhibitor, can inhibit the expression of tumor cell glycoprotein andrestrain the tumor cells from growth and transfer. In addition, it can stimulate body immunologic system and enhance body immunity function, immunocyte generation and the activity of killing tumor cells. Because of its satisfactory antineplastic-activity and immunity-enhancement, swainsonine as the potential medicine of antineoplastic agents screening, is paid great attention to. Though, the study of swainsonine clinical treatment effect has been the phase Ⅲ abroad, it is inchoate in home at present. This article mainly revises the progress in research of swainsonine antineplastic-activity in both the direct and the indirect activity, and introduces the effects of swainsonine as a therapeutic drug.
