Study on chemical composition and structure of a polysaccharide from Monostromaceae sp.
本文对一种礁膜多糖进行化学组成和结构研究。礁膜藻体经热水提取、乙醇沉淀、冻干得礁膜粗多糖,经Q Sepharose Fast Flow和Sephacryl S-400/HR分离纯化,得到多糖组分MPS。高效凝胶渗透色谱法分析表明MPS为纯度高的均一多糖,分子量约为58.4 kDa;气相色谱法测得多糖MPS主要由鼠李糖组成,还含有少量木糖、葡萄糖、甘露糖和半乳糖;MPS硫酸根和糖醛酸含量分别为28.5%和3.94%;经甲基化分析和核磁共振波谱分析表明,MPS主要由→3)-α-Rhap-(1→、→2,3)-α-Rhap-(1→和→2)-α-Rhap-(1→组成,还存在少量的→2,3,4)-α-Rhap-(1→、→2,4)-α-Rhap-(1→、α-Rhap-(1→和→3,4)-α-Rhap-(1→,硫酸基团主要位于→3)-α-Rhap-(1→的C-2位和→2)-α-Rhap-(1→的C-3位。本研究表明多糖MPS是一种结构新颖的硫酸多糖。
polysaccharide MPS was extracted from Monostromaceae sp. with hot water, and further purified by a combination of Q Sepharose Fast Flow column and Sephacryl S-400/HR column. MPS was a homogeneous polysaccharide in size and charge, with an average molecular weight of about 58.4 kDa. Chemical composition analysis indicated that MPS contained 28.5% sulfate ester and 3.94% glucuronic acid. Monosaccharide composition analysis by GC demonstrated that MPS was mainly composed of rhamnose with minor amounts of xylose, glucose, mannose and galactose. On the basis of methylation and NMR spectroscopic analyses, the chain of the polysaccharide MPS was characterized to consist of →3)-α-Rhap-(1→, →2,3)-α-Rhap-(1→ and →2)-α-Rhap-(1→. Minor amounts of →2,3,4)-α-Rhap-(1→, →2,4)-α-Rhap-(1→, α-Rhap-(1→ and →3,4)-α-Rhap-(1→ were also detected. The sulfate groups were substituted at C-2 of the →3)-α-Rhap-(1→ and C-3 of the →2)-α-Rhap-(1→. The investigation demonstrated that MPS was a sulfated polysaccharide with different structural characteristics from other sulfated polysaccharides from seaweeds.
