Jagged-1对Th17细胞分化影响的初探
Effect of Jagged-1 signaling on the differentiation of Th17 cells
用可溶性Jagged-1/Fc嵌合蛋白在体外诱导小鼠淋巴结细胞分化,初步探讨Notch信号通路的活化在Th17细胞分化中的作用。用不同浓度Jagged-1在不同时间刺激小鼠淋巴结细胞,ELISA 检测细胞培养上清中IL-17、IL-4、IL-6 和IL-10的水平;Western blot检测细胞内NICD、Hes-1和Deltex-1蛋白的表达;流式细胞术分析 Jagged-1对CD4+ T细胞胞内IL-17A生成的影响。结果显示,随着Jagged-1浓度的增高,淋巴结细胞的NICD、Hes-1和Deltex-1的表达明显增高,IL-17的表达下降,IL-10的表达升高,而DAPT和anti-Jagged-1能够抑制Jagged-1的这种作用。结果初步提示Jagged-1 能上调Notch信号通路抑制外周T淋巴细胞向Th17细胞分化。
soluble Jagged-1/Fc chimera protein (Jagged-a/Fc) was directly used to induce in vitro differentiation of murine lymph node cells to explore primarily the effect of activation of Notch signaling on the differentiation of Th17 cells. First, the lymph node cells were treated with different concentrations of Jagged-1 at different time. Then, interleukin 17(IL-17), interleukin 4(IL-4), interleukin 6(IL-6) and interleukin 10 (IL-10) in the culture supernatant from the treated cells were detected by ELISA. And expression of NICD, Hes-1 and Deltex-1 was determined through Western Blot, respectively. Flow cytometry was applied to analyze the effect of Jagged-1 on the intracellular level of IL-17 in CD4+ T cells. The results showed that, as the concentration of Jagged-1 was increased, expression of NICD, Hes-1 and Deltex-1 was obviously up-regulated, the level of IL-10 also was increased, whereas expression of IL-17 was decreased, which could be offset by DAPT or anti-Jagged-1 antibody. The results indicate that Jagged-1 could up-regulate Notch signaling, suppressing the differentiation of peripheral T- lymphocytes into Th17 cells.
