HPLC-ELSD法测定浙贝乙素胆酸盐中浙贝乙素和胆酸的含量

目的 建立浙贝乙素胆酸盐中浙贝乙素和胆酸的含量测定方法。 方法 以甲醇-乙酸铵(0.1 moloL-1)-三乙胺-冰乙酸(78:22:0.2:0.03)为流动相,C18柱(250mm×4.6mm,5μm)分离,流速为1 mLomin-1;蒸发光散射检测器检测(漂移管温度72.1℃,氮气流速2 mLomin-1)。结果 浙贝乙素胆酸盐中浙贝乙素和胆酸分别在50.4 ~151.1 μgomL-1(r=0.999 6, n=5)、50.5~151.4 μgomL-1(r=0.999 9, n=5)内峰面积与浓度呈良好线性关系;平均回收率分别为99.19%(RSD=0.62%)和99.37%(RSD=0.63%)。结论 采用本法测定浙贝乙素胆酸盐中的浙贝乙素和胆酸含量,方法简便、可靠、重复性好。
OBJECTIVE To establish a HPLC method for the determination of verticinone and cholic acid in verticinone-cholic acid salt. METHODS The separation was performed on a C18 column (250mm×4.6mm,5μm).The mobile phase was methanol : ammonium acetate (0.1moloL-1) : triethylamine : ethanoic acid (78:22:0.2:0.03). The flow rate was 1mLomin-1. The gas flow rate of ELSD was 2 mLomin-1 and the temperature of drift tube was 72.1℃. RESULTS The linear ranges of verticinone and cholic acid were 50.4 ~151.1μgomL-1 (r=0.999 6, n=5) and 50.5~151.4μgomL-1 (r=0.999 9, n=5) respectively. The average recoveries of the two components were 99.37% (RSD=0.63%, n=9) and 99.19%(RSD=0.62%,n=9) respectively. CONCLUSION The method is simple and reliable for the quality control of verticinone-cholicacid salt.
