褐飞虱是一种迁飞性害虫,为亚洲国家当前水稻上的首要害虫。研究表明flightin在褐飞虱翅型分化过程中扮演着重要的角色。为了探讨不同翅型和龄期褐飞虱体内的flightin蛋白表达情况,本文通过Gateway重组技术构建原核表达载体pDEST17-flightin,以终浓度为1mmol/L的IPTG诱导的蛋白为抗原,免疫新西兰大白兔获得多克隆抗体。Western blot检测表明制备的flightin抗体可特异性地检测褐飞虱体内的flightin蛋白。间接ELISA检测结果表明所制备的抗体效价达1:6400。flightin蛋白在褐飞虱长翅成虫中大量表达,在短翅雄虫中也检测到有微量的表达,而在短翅雌虫和若虫中则检测不到。

he brown planthopper (BPH; Nilaparvata lugens Stal), a kind of migratory insects, is the primary pests of rice in Asian countries. Previous studies on flightin gene indicated that it is playing a crucial role in the process of BPH wing dimorphism. To determine the expression difference of flightin protein in different wing forms and different instar of BPHs, a prokaryotic expression vector pDEST17-flight was constructed through the Gateway recombination technology. Anti-flightin polyclonal antibody was obtained from the antisera of New Zealand white rabbit, which was injected with IPTG-induced protein as antigen. The working concentration of IPTG was 1mmol/L. Western blotting analysis indicated that the flightin antibodies can specifically react with flightin protein in BPH. The ELISA assay showed that the potency of antiserum was 1:6400. Strong expression of flightin protein was detected in macropterous BPH adults and trace expression of flightin protein was detected in brachypterous BPH male ones, but no expression was detected in the brachypterous female ones and nymph ones.
