茶树花粉总RNA提取方法研究
Study on Total RNA Extraction from Camellia sinensis Pollen
以茶树品种'龙井43'的花粉为材料,比较了简易CTAB-LiCl法、CTAB+吸附柱法、Trizol试剂盒提取法和多糖多酚RNA提取试剂盒提取花粉总RNA的效果。结果显示:与前两种方法相比,试剂盒提取的RNA含量较高,质量较好;且多糖多酚RNA提取试剂盒提取的RNA纯度更高,琼脂糖凝胶电泳检测显示,28S和18S条带较清晰且污染少。通过反转录和特异引物扩增实验进一步验证表明,所提的RNA反转录后能扩增出预期的基因片段,可很好的应用于基因克隆与基因表达等分子生物学实验研究。
otal RNA extracted from Camellia sinensis pollen with four methods of CTAB-LiCl, CTAB+adsorption column RNA extraction method, Trizol kit and RNA kit for plant rich in polysaccharides and polyphenol was compared in this study. The results showed that higher yield RNA with good quality was isolated with RNA kits. The electrophoresis assay showed that bands of 28S and 18S were all clear. Further experiments showed that the total RNA could be well specific amplifying of gene fragments after reverse transcription. The results confirmed that the total RNA could be well used for future gene cloning and expression studies.
