甜瓜CmERFV-4基因cDNA克隆及特性分析
Cloning and Characteristic Analysis of CmERFV-4 in Melon(Cucumis melo L.)
乙稀应答因子(ethylene-responsive factor,ERF)是乙稀信号转导途径中的下游组分,可以通过认别并结合靶基因启动子顺式作用元件GCC-box调节多种功能基因的表达,从而实现对植物发育的调控和逆境胁迫的响应。本研究以甜瓜品种河套蜜瓜为研究材料,从成熟甜瓜果实中克隆得到554 bp的CmERFV-4基因cDNA,基于该cDNA序列,进行了蛋白理化性质、空间结构与系统进化分析。实时荧光定量PCR检测结果表明,在不同组织检测结果中CmERFV-4基因在子叶中的表达量最高,在不同发育阶段果实检测结果中该基因在授粉后40d果实中的表达量最高。构建了该基因超表达载体和RNAi载体。?????

In ethylene signal transduction pathway, ethylene-responsive factors (ERF)is the downstream component which can regulate plant development and response to stress by identifying and connecting with the cis element GCC-box of target gene. In this study, CmERFV-4 gene cDNA, 554 bp, was cloned from ripe fruit of melon cultivar Hetao (Cucumis melo L. ). Based on the cDNA sequence, we analyzed the physicochemical properties, spatial structure and phylogenetic analysis of CmERFV-4. Real-time quantitative PCR results showed that the level of expression was highest in the cotyledon in melon with different tissue and in the 40d fruit compared with other fruit development period. The overexpression vector and RNAi vector of this gene were constructed.?????
