急性六价铬中毒对小鼠成体神经发生影响的研究

目的 探讨和研究急性六价铬中毒对成年小鼠海马齿状回神经干细胞增殖和新生细胞存活的影响。方法 雄性成年昆明小鼠分为六价铬染毒组和生理盐水对照组,通过腹腔注射进行六价铬染毒,用BrdU腹腔注射和免疫组化方法定量研究齿状回神经干细胞的增殖和新生细胞的存活状况。结果:染毒组小鼠每1000μm齿状回颗粒层中的BrdU阳性细胞数比对照组增加了17%(P<0.05),而染毒组新生细胞的存活量比对照组明显下降了25%(P<0.05),表明六价铬中毒对小鼠神经干细胞增殖和存活产生了显著影响。结论 急性六价铬中毒促进成年小鼠海马神经干细胞的增殖,但同时引起新生细胞的存活下降,这些变化可能是生物体对六价铬中毒做出的生理应对的细胞基础,最终使自身从铬中毒的损伤中得到恢复。

[Objective]To investigate the influence of acute chromium(Ⅵ)poisoning on the proliferation and survival of adult neural stem cells in the SGZ of the dentate gyrus in mice. [Methods]Healthy Adult Kunming mice were divided into two groups: control group and experimental poisoning group.Exposure to chromium(Ⅵ)by intraperitoneal injection, Immunocytochemistry was used to investigate quantificationally the proliferation and survival of adult neural stem cells in the SGZ of the dentate gyrus in mice.[Results]The number of BrdU-positive cells per 1000μm in the dentate gyrus of experimental poisoning mice was significantly increased by 17% compared with the control group (P<0.05),however the newly generated cells of the survival significantly decreased by 25% in experimental poisoning group compared with the control group (P<0.05),indicating that the differentiation and survival of adult neural stem cells were significant influenced by the acute chromium(Ⅵ)poisoning. [Conclusion]These results suggest that cute chromium(Ⅵ)poisoning enhances the proliferation of adult neural stem cells and induces the deficit in the survival of newly generated cells in the SGZ of the dentate gyrus in mice, the alterations in level of cell is foundation that may help the organism deal with chromium(Ⅵ)poisoning and urge itself to recover from injury.
