利用比较基因组学对QPm.cass-4DL进行精细定位

Fine mapping of adult plant resistance locus QPm.caas-4DL by using comparative genomics analysis

研究表明水稻和短柄草的基因组序列与小麦基因组十分相似,从而利用比较基因组学对小麦功能基因进行精细定位颇为重要。本研究利用小麦、短柄草和水稻的基因组共线性关系对小麦白粉病成株抗性位点QPm.caas-4DL进行比较基因组分析,明确QPm.caas-4DL所在4DL染色体区域与短柄草的1号染色体和水稻基因组的3号染色体直系同源,开发出10对序列标签位点标记,其中与抗病位点QPm.caas-4DL紧密连锁的分子标记为CV6 (0.3 cM). 通过对56份小麦育种品系检测,验证该标记可以直接应用于小麦抗病分子辅助选择。

he genome sequences of rice and Brachypodium distachyon were very similar to wheat genome, which is very important to fine mapping some useful genes in wheat. In the present study, we used comparative genomics analysis between wheat, rice and Brachypodium genomic collinearity to confirm that the powdery mildew resistance locus QPm.caas-4DL in wheat chromosome 4DL was orthologous to Brachypodium chromosome 1 and rice chromosome 3. A total of 10 STS (sequence Tagged site) have been developed and only one STS (named as CV6) has been mapped to the linkage map within 0.3 cM from the resistance locus. This molecular marker can be used in wheat breeding program to bread powdery mildew resistant wheat cultivar by marker assistant selection.
