目的:以牙髓干细胞(DPSCs)为种子细胞,以处理后的牙本质磨片为支架体外构建成牙本质样细胞分化模型,评价该模型对DPSCs定向分化为成牙本质样细胞的有效性。方法: I型胶原酶消化组织块法体外培养DPSCs,鉴定细胞来源和干细胞表面标记;以经过处理的牙本质磨片为支架接种DPSCs,在接种后的第1、2、4、8天分别收集细胞,用扫描电镜和免疫荧光显微镜观察细胞在分化模型中的形态学变化及微丝结构的改变;收集接种后的第1、3、6、8、10天的细胞,行MTT法定量测定DPSCs在分化模型中的增殖活性;免疫组化定量检测成牙本质细胞早期分化标记物碱性磷酸酶(ALP) 的活性变化。结果:在成牙本质样细胞分化模型中,DPSCs在接种早期即形成复层生长,部分细胞可形成成牙本质细胞样形态;其微丝骨架发生重组,增殖活性明显降低,碱性磷酸酶活性增高。提示以牙本质磨片为支架构建的成牙本质样细胞分化模型具有促牙髓干细胞定向分化为成牙本质样细胞的能力。结论:牙本质磨片可作为支架材料构建成牙本质样细胞分化模型,促牙髓干细胞向成牙本质样细胞分化。

o construct odontoblast-like cells’ differentiated models in vitro by inoculated dental pulp cells onto processed dentin discs,evaluate the efficacy of the models . Primary cultures of human dental pulp cells were seeded onto treated dentin discs.For the process of cells’adherence and morphologic change ,samples were collected on day 1,2,4 and 8. Results were analyzed by scanning electron microscope(SEM) and fluorescence microscopy . Samples from the function study were retrieved on day 1,3,6,8 and 10, and evaluated by measuring cell proliferation rate ,alkaline phosphatase (ALP) activity.In the differentiated models, DPSCs can form ambo-layers growth in the earlier period,meanwhile,some cells oriented differentiated into odontoblast-shaped cells. On the dentin discs, the skeleton of microfilaments recombinate(the microfilaments were organized in parallel stress fibre oriented depending on the longitudinal axis of the cytoplasm), obviously step down the DPSCs’ proliferative activity and increase the activity of alkaline phosphatase . The result indicated that the odontoblast-like cells’ differentiated models have the ability to promote the DPSCs differentiating into odontoblasts.The dentin discs can be used to construct odontoblast-like cell’s differentiated models, promoting DPSCs differentiate into odontoblasts.
