甘蔗纤维素合成酶家族的基因组演化
Gene Evolution of Cellulose Synthase Family in Saccharum Spontaneum
应用比较基因组学手段,以甘蔗的近缘物种高粱的11个纤维素合成酶基因家族成员设计杂交探针,从甘蔗割手密的BAC文库鉴定测序得到其中10个甘蔗CesA基因家族成员。另外,通过cDNA克隆得到甘蔗的CesA8。进一步的分析结果表明,甘蔗和高粱的CesA基因家族成员有很高的同源性。用甘蔗割手密CesA家族(SsCesA)与单子叶植物水稻、玉米、高粱及双子叶植物拟南芥、葡萄的CesA基因家族系统进化分析预测,结果推测单双子叶植物的CesA基因可能由6个共同祖先基因起源,其中甘蔗的CesA2基因的演化速率较快。根据模式植物拟南芥和水稻CesA基因关于初生和次生细胞壁形成的特异性与甘蔗CesA基因的聚类关系分析,预测SsCesA1、SsCesA6和SsCesA9基因可能与初生细胞壁形成相关,SsCesA10、SsCesA11和SsCesA12基因对于次生细胞壁形成是必需的。这为后续甘蔗CesA基因家族的功能和分子机制的研究奠定基础。
ccording to the comparative genomics analysis between sorghum and Saccharum species, hybridization probes for eleven cellulose synthase family genes were designed for screening a Saccharum spontaneum BAC library. The results showed that, besides CesA8, ten of orthologous cellulose synthase gene family members in the BAC library were identified and sequenced. The absent orthologous gene, CesA8 were cloned with cDNA template PCR.The analysis of gene annotation and sequence comparison demonstrated that the orthologous gene pairs of sorghum and sugarcane shared high sequence similarity. The phylogenetic genetic analysis were performed for momocotyledon(sugarcane, rice, maize and sorghum) and dicotyledon (Arabidopsis and grape) of cellulose synthase gene family, suggesting that there were at least six ancestor genes for the two classes and SsCesA2 was rapidly evolving compared to the other genes. Based on previous studies for the CesA family in the model plants(Arabidopsis and rice) and phylogenetic genetic classification, SsCesA1, SsCesA6 and SsCesA9 were predicted to play important roles in primary cell wall formation, and SsCesA10, SsCesA11 and SsCesA12 were indispensable for secondary cell wall formation. This study provided the foundation for further investigate the function of CesA family and molecular mechanism of cellulose synthase in sugarcane.
