枸杞叶总黄酮对UVB辐射导致的光损伤无毛小鼠皮肤组织MDA、Hyp含量及SOD、GSH-Px、CAT活力的影响

目的:研究枸杞叶总黄酮对紫外线辐射导致小鼠皮肤光损伤的作用及其机理。方法:将60只昆明种小鼠剃除背部毛随机分成6组,除空白对照组外,模拟UVB照射2h/天,连续照射两周,紫外模型组直接照射UVB,基质组照射前涂抹等剂量乳膏基质,给药组照射前分别在裸露背部皮肤处外用高、中、低剂量的枸杞总黄酮乳膏。测定最小红斑量(MED),光镜观察皮肤结构组织学改变,测定皮肤组织MDA、Hyp含量以及SOD、GSH-Px和CAT活力。结果:两周后,模型组小鼠皮肤组织MDA含量增高,Hyp含量降低,SOD、GSH-Px和CAT 活性明显降低(P<0.01),病理切片呈现光老化状态,有炎症反应;与模型组比较,枸杞叶总黄酮高剂量组最小红斑量增多(P<0.01),SPF=20,皮肤组织病理切片可见光损伤状态减轻,并且可显著降低MDA、增高Hyp含量,提高SOD、GSH-Px、CAT 活性(P<0.01)。结论:外用枸杞叶总黄酮可防止UVB辐射导致的皮肤损伤。
Objective: To study the influence of topical using total flavonoids from Lycium barbarum leaves cream on the skin-photodamaged caused by UVB radiation. Methods: 60 hairless mice were randomly divided into 6 groups, in addition to the control group, with UVB irradiation 2h/day for two weeks. UVB direct exposure to UVB model group. Before the irradiation, the base group smear cream base. And the treatment groups were irradiated to dorsal skin after topical using low, medium and high doses of total flavonoids from Lycium barbarum leaves cream. Determinating minimal erythema dose(MED), histological changes of skin tissue structure, the activities of SOD, GSH-Px, CAT, and the content of MDA and Hyp in skin tissue. Results: After 14 days, MDA content of model group increased, Hyp content decreased, the activities of SOD, GSH-Px, CAT decreased significantly (P<0.01), mice skin tissue histology showing photoaging state and inflammation. Compared with the model group, the MED of Lycium barbarum leaves flavonoids high-dose group was significantly increased (P<0.01), and the SPF=20, the photodamage was midigated, the content of MDA in dorsal skin tissue was significantly reduced, Hyp content was increased, the activities of SOD, GSH-Px and CAT were increased(P<0.01). Conclusion: Topical using total flavonoids from Lycium barbarum leaves can prevent skin damage caused by UVB radiation.
