不同剂量TGF-β1对颞下颌关节炎软骨下骨的影响
Effect of different dose of TGF-β1 on subchondral bone in osteoarthritic TMJ
目的 探讨关节腔内注射TGF-β1治疗颞下颌关节骨关节炎(TMJOA)的效果。方法36只新西兰大白兔随机分为三组,全部采用双侧关节盘部分切除法建立TMJOA动物模型,建模四周以后三组动物分别采用右侧关节腔内注射20, 40, 和 80 ng TGF-β1。对照侧关节腔内注射等量生理盐水另取四只动物作为正常对照组。关节腔注射药物后分别于6周、12周每组处死6只动物,解剖颞下颌关节,进行组织学以及microCT检测。结果 所有注射TGF-β1组其髁突关节软骨都好于较盐水对照组,12周时要好于6周组标本。所有注射TGF-β1组软骨下骨微结构均与正常组无明显差异。而对照组呈现典型的颞下颌关节骨关节炎软骨下骨硬化的病理表现。结论 关节腔内注射TGF-β1可以有效预防颞下颌关节骨关节炎软骨下骨病变的发生。
Objective This study sought to evaluate the effects of intra-articular injection of different dose of TGF-β1 on the subchondral cancellous bone repair in osteoarthritis (OA) of temporomandibular joint (TMJ). Methods Partial disc perforation was performed bilaterally to induce an osteoarthritic joint in 36 animals. 20, 40, or 80 ng TGF-β1 were injected into the right joint, and the contralateral joint received vehicle injection. 4 additional animals were used as normal control. The animals were sacrificed at 6 and 12 weeks after the first injection, respectively, for histology and micro-CT examinations.Micro-CT scanned to quantify three-dimensional microarchitecture of subchondral bone. Results All TGF-β1-treated joints were covered by a layer of well-organized fibrocartilage, and had normal mictoarthitectural properties, whereas the vehicle-treated joint had typical OA-related cartilage degradation and subchondral bone sclerosis.. Conclusion These results suggested that TGF-β1 administration is an effective therapy for intervention of subchondral bone lesions in TMJ OA.
