长角血蜱微生物群落及其共生菌的研究
Microbial communities and symbionts in the hard tick Haemaphysalis longicornis (Acari: Ixodidae)
蜱与体内微生物群落的密切关系,对蜱的适应性和病原菌的定植及传播具有重要作用。长角血蜱(Haemaphysalis longicornis)属三宿主蜱,在我国广泛分布。该蜱可传播多种病原菌而导致疾病,对公共卫生和畜牧业造成严重危害。本文以长角血蜱为对象,运用分子生物学方法对其体内微生物群落的组成和共生菌进行了分析。结果显示:长角血蜱野外种群体内微生物具有较丰富的多样性,并检测到三类共生菌类考克次氏体(CLS-Hl和NCLS-Hl)、类杀雄菌(ALS-Hl)和类立克次氏体(RLS-Hl)共存于长角血蜱体内。CLS-Hl感染特定的组织,卵巢和马氏管;ALS-Hl和RLS-Hl不存在感染特异性,在卵巢、唾液腺、马氏管和中肠中均存在。感染率检测显示,在长角血蜱野外种群中,CLS-Hl的感染率为100%,ALS-Hl在雌蜱和雄蜱中的感染率分别为33.3%和83.3%;RLS-Hl在雌蜱和雄蜱中的感染率分别为88.9%和100%。在实验室种群中,只有CLS-Hl可稳定传递,推测CLS-Hl为长角血蜱的专性共生菌。本研究揭示了宿主蜱与微生物之间复杂的生态相互作用,为开展蜱类有效防治提供思路。
lose relationships between ticks and microbial communities are important for tick fitness and pathogen colonization and transmission. Haemaphysalis longicornis, a three-host tick, distributed widely in China, can carry and transmit various pathogens and pose serious damages to public health and livestock. In the present study, we investigated the composition of bacterial communities associated with H. longicornis and evaluated the putative symbionts using molecular biology methods. The results showed that diverse microbial communities had been found from the field-collected H. longicornis. Three types of symbionts Coxiella-like(CLS-Hl和NCLS-Hl), Arsenophonus-like(ALS-Hl)and Rickettsia-like symbionts(RLS-Hl)were identified in a single host simultaneously. In females, CLS-Hl colonized specifically in ovaries and malpighian tubes, while ALS-Hl and RLS-Hl infected all tissues tested (ovaries, salivary glands, malpighian tubes and midguts). In natural field population, CLS-Hl showed 100% infection rate; ALS-Hl occurred in 33.3% females and 83.3% males; the prevalence of RLS-Hl was 88.9% and 100% in females and males, respectively. However, only Coxiella-like bacteria were transmitted stably in the laboratory-reared ticks. We speculate that an obligate symbiotic association between Coxiella symbiont and its host. Our investigation of microbial communities in the ticks revealed the complexity of ecological interactions between host and microbe and provided insight for the biological control of ticks.
