黄瓜花叶病毒2b 蛋白关键氨基酸的致病性分析

FRad352b-CMV 是用Rad35-CMV 株系的2b 基因精确替换Fny-CMV 株系的2b 基因 构建而成,其症状表现为轻花叶。本研究发现了FRad352b-CMV 的重花叶症状突变体 FRad35Leu2b-CMV。Northern 杂交和序列分析结果表明:2b 蛋白第55 位 Leu 对2b 蛋白行使 致病功能具有关键的作用。在已诱导绿色荧光蛋白(GFP)系统沉默的转GFP 本生烟16c 上分 别接种FRad35Leu2b-CMV 和FRad352b-CMV,结果证明Leu55 在FRad35Leu2b-CMV 在抑制寄 主RNA 转录后基因沉默(PTGS)的过程中起关键作用。构建FRad35Leu2b-CMV 和 FRad352b-CMV 的2b 蛋白的农杆菌瞬时表达载体35S-2bLeu 和35S-2bPro,并通过农杆菌将其 与35S-GFP 共浸润转GFP 本生烟16c 叶组织分别瞬时表达2bLeu 和2bPro。从对浸润后第3 天、第7 天、第14 天叶组织中GFP mRNA 和GFP siRNA 的分析结果,我们推测2bLeu 介导 的高致病性是通过阻止siRNA 的系统移动,使得病毒更为有效地系统移动和复制积累,导致寄主表现严重的病毒症状。
MV recombinant FRad352b-CMV is generated by replacing 2b gene in Fny-CMV with 2b gene of Rad35-CMV, inducing mild mosaic. In this study, we found FRad352b-CMV severe mosaic symptom mutation of FRad35Leu2b-CMV. Northern blotting and sequence analysis showed: the Leu55 of FRad352bLeu-CMV 2b protein plays an important role in the pathogenicity mediated by the 2b protein. we inoculated FRad35Leu2b-CMV and FRad352b-CMV on GFP-transgenic N. benthamiana plant (line 16c), in which GFP was pre-silenced by by infiltration with a hypervirulent strain of A.tumefaciens carrying the 35S-GFP plasmid. It is concluded that Leu55 plays a critical role in suppressing PTGS. we constructed agrobacteria transient expression vectors of 2b gene of FRad35Leu2b-CMV, FRad35Leu2b-CMV, 35S-2bPro and 35S-2bLeu. When agrobacteria carrying these vectors co-infiltrated with that carrying 35S-GFP on Nicotiana benthamiana plant (line 16c), northern blot analyzes accumulation of GFP mRNA and GFP siRNA in the tested plants at 3d丆7d丆and 14d. We speculate that high virulence mediated by 2bLeu is achieved through blocking systemic movement of viral siRNA, making CMV systemically move and replicate more effectively, then resulting to induction of severe viral symptom on hosts.
