利用大肠杆菌和金黄色葡萄球菌混合液诱导家蝇幼虫,抽取血淋巴。将无细胞的血淋巴酸化以后用Sep-Pak C18柱进行固相萃取,分别以10%、30%和80%浓度的乙腈(ACN)分步洗脱,将各洗脱组分冻干后分别测定抗菌活性。对10%和30%的固相萃取组分利用反向高效液相色谱(RP-HPLC)进行了分离纯化,收集各峰,并利用液体生长抑制法检测其抑菌活性,发现10%组分和30%组分中分别有三个峰对大肠杆菌有很强的活性,10%组分有两个峰对金黄色葡萄球菌有明显的抑制活性,而30%组分对金黄色葡萄球菌无明显的抑制活性。

Hemolymph extracted from Musca domestica larvae was treated with the mixtrue of living Escherichia coli and Staphylococcus aureus。 After acidification with trifluoroacetic acide, the hemolymph was fractionated on Sep-Pak C18 column with 10%, 30% and 80% of acetonitrile in succession and the crude components were concentrated by vacuum centrifuge, and then their antibacteiral activities were monitored with the method of cylinder-plate。 Further purification was conducted on crude components seperated with 10% and 30% of ACN through RP-HPLC, and antimicrobial activities of sub-components were measured with liquid growth inhibition method。 The results show that three and two sub-components in the crude component from 10% ACN of extract had high activity against Escherichia coli and Staphylococcus aureus respectively, and three sub-components from 30% of extract had distinguishable activity against Escherichia coli.
