目的:研究体外条件下3-羟基丁酸(3-HB)对神经干细胞(NSCs)增殖及分化的影响。方法:分离孕14.5天大鼠胚胎大脑皮质NSCs,进行原代培养;培养至第三代的NSCs分别用0.1、0.05、0.02、0.01g/l 3-HB处理,未加药组作为对照,CCK-8试剂盒检测细胞增殖、活力,免疫细胞荧光化学观察NSCs分化情况。结果:与对照组相比,3-HB处理对NSCs的增殖、活力没有显著影响;3-HB处理组β-tubulin阳性细胞(神经元)比例增加,GFAP阳性细胞(胶质细胞)比例没有显著变化。结论:3-HB对分离培养的NSCs在体外的增殖和活力无明显影响,但能够促进其向神经元方向分化。

Objective: Learn the effect of 3-hydroxybutyrate(3-HB) on the viability and differentiation of neural stem cell in vitro. Method: NSCs were isolated from the cerebral cortex of rat embryos on embryonic day 14-15, and P3 cells were used; NSCs were treated with different concentration(0.1、0.05、0.02、0.01g/l) of 3-HB, untreated as control. Cell viability was detected by CCK-8, and immune cell fluorescence chemistry was used to study the differentiation of NSCs. Result: Cell viability had no significant difference between 3-HB treated group and control. The percentage of β-tubulin positive cells was increased, but the percentage of GFAP positive cells had no dramatically alteration. Conclusion: Different concentration of 3-HB had no significant influence on NSCs viability, but could promote NSCs differentiate into neurons.
