Gamma氨基丁酸(GABA)参与了包括肿瘤细胞在内的多种细胞的增殖、分化和迁移的过程。但是GABA在小儿急性淋巴细胞白血病(acute lymphoblastic leukemia,ALL)的作用尚不明确。本次实验的目的是探讨GABA能信号系统在ALL患儿的骨髓淋巴细胞中的表达情况及其对白血病细胞增殖的影响。应用RT-RCR、Western Blot以及免疫荧光染色法发现,在19位ALL患儿的骨髓淋巴细胞中,谷氨酸脱羧酶(GAD65/67)和A型GABA受体(GABAARs)的亚单位均有表达。但是应用MTT检测法发现,GABA或GABAARs特异性激动剂muscimol对体外培养的Jurkat淋巴细胞的增殖没有明显的影响,提示骨髓淋巴细胞中的GABA能信号系统可能不参与ALL患儿骨髓白血病细胞增殖的病理过程。

Gamma-aminobutyric acid (GABA) is involved in the proliferation, differentiation, and migration of several cell types including cancer cells. Whether GABA may be involved with acute lymphoblastic leukemia (ALL) is unclear. Therefore, the goal of this report was to examine if GABAergic signaling is expressed in bone marrow lymphocytes of ALL children. RT-PCR, Western blot, and immunostaining assays revealed that the GABA synthetizing enzyme glutamic acid decarboxylase (GAD) isoforms (GAD65 and GAD67) and GABAAR subunits were expressed in bone marrow lymphocytes of ALL children. However, treatment with GABA or selective GABAAR agonist muscimol did not affect the proliferation of cultured Jukat cells. Thus, the role of GABAergic signaling system in the progression of ALL requires further investigation.
