本文利用胶原/壳聚糖支架为载体,模拟体液环境研究肝细胞生长因子对大鼠骨髓间充质干细胞(BMSC)向毛囊细胞分化的影响。通过注射法将大鼠骨髓间充质干细胞植入胶原/壳聚糖支架, 考察加入外源性的肝细胞生长因子对BMSC向毛囊细胞分化的影响。利用HE染色技术观察了支架中细胞形态,利用MTT法评价了支架的细胞毒性。采用逆转录聚合酶反应(RT-PCR)以及Western印迹法(Western Blotting, WB)检测细胞分化的相关指标。经过三周的诱导,毛囊细胞的特征标志物在基因水平和蛋白水平上都呈现高表达。 实验结果表明,胶原/壳聚糖支架利于骨髓间充质干细胞的增殖,加入外源性肝细胞生长因子能促进骨髓间充质干细胞往毛囊细胞的分化。

iming to study the effect of Hepatocyte growth factor on the differentiation of bone mesenchymal stem cells(BMSC) into hair follicle cells, collagen/chitosan scaffold was chosen as carrier to estimate the in vivo environment. MSC were injected into scaffold and induction started by immersing scaffolds in low serum medium with HGF after cells proliferating for 5 days. Methods of cell staining and histological section were used to investigate cell morphology. Quantitative real time-polymerase chain reaction (RT-PCR) analysis and Western Blotting (WB) were used to detect key markers of hair follicle celss both in gene and protein level. Three weeks after induction, key markers were highly expressed compared with normal MSC. All the results showed the biocompatibility of collagen/chitosan scaffolds and active effect of HGF on the differentiation of MSC into hair follicle cells.
