微生物的分子生物学鉴定方法
he molecular methods for identification of hydrophytic microbes
传统的微生物分类和鉴定方法主要是以微生物的形态学和生理生化等特性为依据,繁琐且费时。近年分子生物学的发展,为微生物的分类鉴定工作提供了较简便准确的方法。分子生物学方法一部分用到PCR扩增,比如:变性和温度梯度凝胶电泳,单链构建多态性,限制性片段长度多态性,自动核糖体间隔基因分析等。一些不依赖PCR的方法也得到广泛的应用,如荧光原位杂交,DNA联合分析等。本文重点综述了几种广泛应用的分子生物学技术并对其原理及优缺点进行了阐述。
he classification and identification of the microorganisms is an important way to the study of microorganism. Traditionally, it is tedious and time-consuming because it follows the character of bioecology .With the development of molecular biology, the way of microbic identification becomes more precise. Most of the molecular biology methods are involved in PCR, for instants, temperature gradient gel electrophoresis, denaturing gradient gel electrophoresis, random sequencing in clone libraries, single-strand conformation polymorphism, terminal-restriction fragment length polymorphism, and so on. Some methods which are not involved in PCR are also be used widely, such as fluorescence in-situ hybridization, DNA re-association analysis. On the whole, we can get different information from different methods. In this paper, we introduce some useful methods and show their advantages and disadvantages.
