簇毛麦高分子量麦谷蛋白亚基(HMW-GS)已被证实对面包品质改良具有显著正效应,但将簇毛麦HMW-GS亚基通过小片段易位系方式转入小麦背景的研究还未见报道。本研究通过电离辐射普通小麦-簇毛麦1V异附加系,从辐射后代中,选育出一个小片段易位系NAU1V-3-5,综合利用染色体C-分带、GISH、双色原位杂交及1V染色体特异分子标记鉴定结果表明,该易位系的易位染色体为T1VS-6BSo6BL。利用SDS-PAGE对易位系进行了HMW-GS亚基组成分析,簇毛麦编码HMW-GS亚基位于中国春7亚基和8亚基之间,为V71亚基,同时易位系NAU1V-3-5也携带V71亚基,表明该亚基基因位于簇毛麦1VS上。携带簇毛麦HMW-GS亚基的易位系NAU1V-3-5为普通小麦品质改良提供了新的种质资源。

Glu-V1 locus, located in the chromosome 1V of Dasypyrum villosum, was proved to have positive effects on grain quality. However, there are very few reports about the transfer of the D. villosum-derived HMW-GS gene into wheat background by chromosome manipulation. In the present study, a small alien-segment translocation line NAU1V-3-5 was developed through ionizing radiation of the mature female gametes of CS - D. villosum 1V disomic addition line and confirmed by cytogenetic analysis. Genomic in situ hybridization, chromosome C-banding and 1V-specific EST-STS markers enabled the translocated chromosome was T1VS-6BSo6BL. And seed storage protein analysis confirmed the Glu-V1 allele of D. villosum was Glu-V1a and its coded protein was V71 subunit. The T1V#4S-6BSo6BL translocation line could be useful germplasms for bread wheat improvement. The developed 1VS-specific molecular markers could be used to rapidly identify and trace the alien chromatin of 1VS in wheat background.
