miR-200b靶向作用ZEB2调控口腔鳞状细胞癌的迁移和侵袭

Purpose：To study the relationship between miR-200b and ZEB2 and the regulation of biological function of them for OSCC cells in vitro experiments.Methods: The expression of miR-200b and ZEB2 in oral cancer cell lines (Tca-8113, SCC-15, Cal-27) were measured, and the dual luciferase reporter system was used to confirm the relationship between miR-200b and ZEB2. In addition, MTT, wound healing assay and Transwell chambers experiment detect the cell proliferation, migration and invasion ability.Results: Compared with the normal oral mucosa cells, the expression of miR-200b in different OSCC cell lines were decreased (P < 0.05), while the mRNA and protein levels of ZEB2 were significantly higher in cancer cells (P < 0.05). pre-miR-200b overexpression can inhibit the Tca-8113 cell proliferation activity, wound healing ability, migration and invasion. The dual luciferase reporter assay and Western Blot assay confirmed that ZEB2 could be targeted by miR-200b. si-ZEB2 can significantly reduce the migration and invasion cell number through the membrane in vitro. Western Blot assay confirmed that the protein expression level of transfecting anti-miR-200b and ZEB2 was higher than that of the only si-ZEB2 transfection group.Conclusion: Our ?ndings suggest that miR-200b can target the 3 '-UTR of ZEB2, and significantly inhibit migration and invasion of OSCC cells.
