N-myc转录调控长链非编码RNA-ncRAN的作用机制研究
N-myc induced the long non-coding RNA, ncRAN, in neuroblastoma
目的 利用生物信息学和分子生物学方法揭示在神经母细胞瘤中转录因子N-myc调控长链非编码RNA-ncRAN的机制。方法 通过内源性沉默或可控性外源表达N-myc,双荧光素酶报告基因分析法,及体外细胞增殖、存活实验,全面系统地证明N-myc与ncRAN的转录调控关系,并探索ncRAN在N-myc所介导的信号通路中的重要作用。 结果 结合生物信息学分析结果及体外实验初步鉴定了位于ncRAN启动子区上游407bp~395bp区域的转录因子N-myc结合位点;细胞增殖及克隆形成实验均提示ncRAN协同促进了N-myc的癌基因功能。结论 ncRAN可能作为N-myc的直接靶基因,参与了N-myc介导的神经母细胞瘤恶性转化及发展。
Object To investigate the mechanism of the long non-coding RNA, ncRAN, induced by N-myc in neuroblastoma with bio-informatics and molecular biology methods. Methods The transcriptional regulation between N-myc and ncRAN was identified by RT-PCR, gene silicing, and dual-luciferase reporter analysis. Cell proliferation assay and Colony formation assay were used to investigate whether ncRAN played an important role in N-myc related cell signal pathway. Results The binding sites of N-myc located on ncRAN promoter of 407bp~395bp which was upstream from start code. The cell proliferation assay and Colony formation assay suggested that ncRAN promoted the carcinogenesis and survival function of N-myc. Conclusion ncRAN as a directed target gene of N-myc envolved in malignant transformation and progression of neuroblastoma.
